I have collected Sticklebacks several times before in all kind of habitats. Salt, brackish and purely fresh. I was just wondering if certain types of Sticklebacks( two, three, four spined etc... ) prefer certain habitats/salinity?

I've been looking into sticklebacks lately. From what I have found, they seem to be the world's most adaptable fish (deliberate exageration for emphasis), at least for water quality parameters. My interest was piqued by reading up on the invasive sticklebacks in the Great Lakes, the three spined and four spined. Some of the three spined and all of the four spined seem to have come in with water balast from ocean going ships (big problem here), so they were marine fish that adapted to fresh water pretty quickly and well. Since these are both established in Lake Superior, they apparently don't require salt to breed.

If you are collecting, take a hydrometer along and check the water at the location. Match that in the aquarium and I would bet you could slowly change them to fresh over time.

Not really an answer to your question, but its what I have learned over the winter. Still more study to do.
